History

Department of physiology of industrial microorganisms was created in 1929. During this time, the department was managed by PhD M.L. Nepomnyashcha (1929-1952), PhD. H.M. Frenkel (1952-1959). From 1960 to 1988, the department was headed by a corresponding member of the Academy of Sciences of Ukraine, Professor E.I. Kvasnikov is a famous scientist, talented teacher and organizer of biological science.

COLLECTIONS OF MICROORGANISMS

The laboratories of the department are the curators of sub-collections of yeast, lactic acid bacteria and actinobacteria, which are included in the Ukrainian collection of microorganisms (UKM) of the Institute of Microbiology and Virology named after D.K. Zabolotny National Academy of Sciences of Ukraine. According to the Resolution of the Cabinet of Ministers of Ukraine No. 527 dated 04/01/1999 The collection of the Institute is included in the objects of the National Heritage of Ukraine.

DEPOSITORY OF MICROORGANISMS

The Depository of Microorganisms was created on the basis of the department, which is the only one in Ukraine that guarantees the state storage of non-pathogenic innovative microorganisms in accordance with the resolution of the Cabinet of Ministers of Ukraine "On the State System of Depositing Microorganism Strains" (No. 705 dated 12.10.94).

Since 2002, dozens of students from different countries of the world have been interning in the department as part of the IAESTE Ukraine program (http://www.iaeste.org.ua)

CURRENT RESEARCH

1. Physiology and probiotic properties of lactic acid bacteria and bifidobacteria. For the first time, ecological approaches in the taxonomy of lactic acid bacteria using taxonomic and molecular genetic methods of identification were proposed. The effect of the isolation medium on the physiological and biochemical properties of lactobacilli, enterococci, lactococci, and bifidobacteria was revealed.

   Universal primers for their genomic DNA have been developed. Confirmed heterogeneity of the 16S rRNA gene of some species of lactic acid bacteria and bifidobacteria. An express system for identification of industrially important strains was created on the basis of PCR. A lactobacillus strain has been identified that produces a specific bacteriocin-like substance encoded by chromosomal genes. On the basis of PCR analysis with primers specific to the three most common genes, the presence of enterocins A, B and P and their expression in probiotic strains of enterococci was revealed. PCR screening among a collection of bifidobacteria revealed the presence of the β-fructofuranosidase gene in some strains, which allows the use of fructooligosaccharides as the only source of carbohydrate nutrition.
   Teichoic acids were obtained from whole cells of industrially important lactobacillus strains using a modified method of their extraction. The composition of teichoic acids and the structural differences of the studied polymers were determined by the methods of infrared spectroscopy and nuclear magnetic resonance. The adhesive activity of lactic acid bacteria and bifidobacteria to human buccal and vaginal epitheliocytes was studied. It is shown that treatment of the buccal epithelium with lactobacillus teichoic acids reduces their adhesion. A dose-dependent stimulating effect of teichoic acids of probiotic lactobacillus strains on the immune system of laboratory animals was established.
   The identified adhesive activity of lactic acid bacteria and bifidobacteria does not depend on the degree of hydrophobicity of the bacterial cell surface and their autoaggregation.
    A collection of strains of lactic acid bacteria and bifidobacteria isolated from different econiches - the digestive tract of people of different age groups (children, adults, long-lived), lactobacilli isolated from women with osteoporosis - was created. The collection includes species living in the body of cold-blooded and warm-blooded animals (mammals, birds, fish, dolphins, bees), as well as isolated from epiphytic microflora and traditional fermented milk products. Selected probiotic strains for functional food products.
    Selection of optimal cultivation conditions for increasing the production of individual exometabolites was carried out, and approaches to their isolation and purification were worked out. New approaches to the synthesis of nanocomposites using metal nanoparticles of exopolysaccharides produced by biologically active strains of lactic acid bacteria of different taxonomic groups as a reducing agent and stabilizer have been developed.

2. Scientific basis of microbial bioremediation.

Ecological and taxonomic studies of hydrocarbon-oxidizing strains of actinobacteria, bioremediation of environments contaminated with oil and oil products, detoxification of aromatic pharmapollutants.

   Based on the study of the biotechnological potential of hydrocarbon-oxidizing actinobacteria collected and isolated from technogenically polluted econiches, the most active strains were selected - destroyers of oil and oil products, which belong to the species Dietzia marisGordonia rubripertinctа and Rhodococcus erythropolis. Their ability to destroy hydrocarbons (in particular, the most difficult oxidizing fractions of oil - petroleum oils and various brands of mineral motor oils) in microaerobic conditions, a wide range of pH, temperature and mineralization of the environment was established. It was shown that the sensitivity of actinobacteria to high concentrations of motor oils correlates with the presence and amount of zinc dithiophosphate in them, the main toxic component of these lubricants. Among the heavy metals included in the composition of the oils, copper, aluminum and zinc ions were found to be the most toxic for the studied strains. It was established that the main mechanisms of adaptation of actinobacteria to assimilation of hydrocarbons are a significant increase in the hydrophobicity of the cell wall and the synthesis of biogenic surfactants (biosurfactants), which exhibit emulsifying properties and form stable emulsions of the "oil/water" type. It was found that the composition of biosurfactants of actinobacteria is dominated by trehalose monomycolates and dimycolates, which are the main glycolipid components of the cell walls of the studied microorganisms. On the basis of the most active hydrocarbon-oxidizing strains of actinobacteria immobilized on an oil-absorbing sorbent, a highly effective ecologically safe biological preparation "Ekolan-M" was created for cleaning the environment from oil and oil products.
  The ability of hydrocarbon-oxidizing strains of actinobacteria to biotransform pharmaceutical waste containing phenolic hydroxyl was revealed, active strains that destroy acetaminophen were selected, the ways of its biotransformation, intermediate and final products of biodestruction were determined, and their environmental safety was established.

    Bacterial transformation of inorganic xenobiotics (metals and non-metals), processes of bioleaching and biosorption. The relevance of the use of microbial remediation of metal-containing effluents, for example, electroplating, is constantly increasing, because the cost of cleaning in this way is 48 times lower than chemical, electrochemical - 73 times, ion exchange - 112 times times, hyperultrafiltration – 187 times. For some chemical compounds (for example, oxygen compounds of chlorine, which are widely used in more than 150 industries), the biological method is the only possible method of neutralization. Salts of perchloric and perchloric acids - chlorates and perchlorates, chromates and compounds of other heavy metals are also the main toxic components of solid rocket fuel and other explosive and incendiary mixtures.
   The work carried out showed that bacteria that neutralize inorganic xenobiotics are widespread in natural and man-made substrates, regardless of the level of their contamination. Active strains of chromate, chlorate, and perchlorate reducers were selected. The most active strains were assigned to species Aerococcus dechloraticansAcinetobacter thermotoleranticusAeromonas dechromaticaPseudomonas putida, optimized growing conditions (carbon sources, pH, Eh, t◦), as well as conditions for maximum neutralization of inorganic oxygen-containing anions, the physiology of the growth of these microorganisms in batch and flow modes of cultivation was investigated. This made it possible to create industrial technologies for neutralizing these pollutants.
   Bacteria were also isolated, which can significantly increase the concentration of rare earth metals in processed ore, absorb compounds of radioactive elements, copper, silver from the environment. Further research could become the basis of a domestic biotech mining industry.

Mycoremediation of the environment from metal pollutants. The department has many years of research experience in the field of geomycology, which is focused on the biogeochemical activity of mycelial fungi and yeasts and the transformation of metals and minerals by fungi in the mycorrhizosphere and lithosphere in natural and technogenic conditions with the aim of their application for mycoremediation. On the basis of these studies, fundamentally new environmental protection biotechnologies were developed for bioremediation of the environment from toxic metals and radionuclides: the use of combined biomineral sorbents based on micromycetes and clay minerals and a method of decontamination of the surfaces of cement barrier building materials from metallic radioactive pollutants (WO 2011121291 A1).

3. Bacillary lectins and their use as effective antiviral drugs. Comprehensive study of lectins of saprophytic microorganisms, to a greater extent extracellular lectins form bacteria of the genus Bаcillus, made it possible to establish the physiological features of the growth of producers and the formation of lectins by them, to carry out directed synthesis of these substances. The technology for obtaining preparations of lectins and their isoforms has been developed. The carbohydrate-binding, hemagglutinating, physico-chemical and medical-biological properties of these biopolymers have been clarified. One of the basic characteristics of these lectins is their affinity for sialic acids. They are able to recognize not only the amount of sialic acids, their types, forms and connections, but also certain sialic oligosaccharide structures on the surface of cells. Sialospecific lectins of bacilli act using different mechanisms. The direct mechanism of action of these lectins is based on blocking the surface sialic receptors of cells and viruses. The indirect mechanism of their action is mediated by cell membrane receptors and leads to the activation of the body's natural protective functions. Different isoforms of bacillus lectins have a regulatory effect at the intracellular level, where the processes of repair, replication and transcription can serve as their targets. Such a complex mechanism of action of bacillary lectins serves as the basis of a wide spectrum of their biological activity. They are inducers of alpha- and gamma-interferon synthesis, exhibit pronounced immunomodulatory, antitumor and antiviral properties. Lectins completely block the surface sialic receptors of influenza, herpes, hepatitis C and HIV viruses, which play a key role in the recognition by viruses of target cells and the initiation of the infectious process, and thus prevent their further adsorption, reproduction and the occurrence of the disease.

   Possible areas of application of bacillus lectins as valuable chemical-analytical and diagnostic reagents and therapeutic and preventive means are proposed. The technology for obtaining gamma-interferon for the prevention and treatment of farm animals using bacterial lectin as an inducer of its synthesis has been developed. Construction of complex preparations of bacillary lectins with heterocyclic bis-adducts as original antitumor drugs was carried out. Based on the lectin of a saprophytic strain B. subtilis IMV B-7014, the drug "LECTIVIR" was created for the treatment and prevention of infections caused by herpes viruses of types 1 and 2. The substance of bacterial lectin and the drug "LECTIVIR" are registered by the Ministry of Health of Ukraine.

4. The influence of non-ionizing electromagnetic radiation of the radio frequency range and geoheliocosmic factors on model microorganisms

     On the yeast Saccharomyces cerevisiae and Shizosaccharomyces pombe, which are recognized models for the study of eukaryotic organisms, it was established that under the influence of non-ionizing electromagnetic radiation of the radio frequency range, changes are observed at the population, cellular and molecular genetic levels. In particular, after irradiation, the effect of non-specific resistance of cells, including to antibiotics, was noted. Assumptions are made regarding the possible role of anthropogenic non-ionizing electromagnetic radiation in the growth of the phenomenon of antibiotic resistance.
   In co-authorship with scientists of the Institute of Radiophysics and Electronics of the National Academy of Sciences of Ukraine, an experimental sample of a biosensor device for assessing the biological effect of non-ionizing electromagnetic radiation was developed and created. The possibility of using luminescent bacteria is being studied Photobacterium phosphoreum in the biosensor system for detecting the biological effect of electromagnetic radiation in the radio frequency range.
     Physiological and biochemical characteristics of microorganisms are being studied under conditions of changing space weather. Particular attention is paid to the participation of inorganic polyphosphates in the reception of helio- and geophysical signals. A unique database of daily monitoring of the metachromasia reaction of polyphosphate-containing volutin granules has been created S. cerevisiae since 2002, which indicates the activity of microorganisms in different periods of solar activity.  

 5. Biological properties, taxonomy, ecology and practical application of yeast.

    The department is traditionally a leader in the country in the field of biology, ecology, taxonomy and practical application of yeast. Currently, research into the biodiversity and biological properties of yeast isolated from authentic Ukrainian food products and other sources is ongoing, with the aim of finding yeast cultures with probiotic potential and producers of useful compounds and products. More than 100 yeast cultures were isolated from home-made dairy products and the gastrointestinal tract of long-lived Carpathians, about 20% of which show antagonistic activity against bacterial pathogens of human diseases. Identified strains - producers of natural flavoring - 2-phenylethanol - among yeast genera Saccharomyces and Kluyveromyces. The maximum level of 2-phenylethanol production in yeast has been established. Found active strains - sorbents of copper, lead and hexavalent chromium ions, which belong to Rhodotorula mucilaginosa, R. aurantiaca, Williopsis californicaCandida kruseiSaccharomyces cerevisiaeCryptococcus sp. It was found that these strains are resistant to high concentrations of heavy metal ions (up to 500 mg/l) in the solution.
    160 yeast strains were isolated from the surface of flowers of various flowering plants. Most of the isolated yeasts showed osmotolerant properties, lipolytic activity, ability to assimilate sorbose and surface activity. Yeast strains Candida gropengiesseri 79a and Candida bombicola 156a synthesized biosurfactants of glycolipid nature, which showed weak antimicrobial activity against gram-positive bacteria
