Areas of scientific work of the department

  • Determination of the fundamental foundations of the physiological role of the interferon system and other immunoregulatory cytokines in normal and pathological processes (infectious-inflammatory diseases of bacterial, viral or mixed viral-bacterial origin, as well as in precancerous and tumorous diseases induced by carcinogenic viruses).
  • The development of scientifically based approaches to the optimization of technologies for obtaining new highly effective recombinant interferon drugs, as well as interferonogenic and antiviral drugs, created on the basis of derivatives and analogs of amixin and molecular complexes of single-chain RNA in combination with monomeric synthetic intercalators, lectins, etc.
  • Development of fundamentally new science-based approaches to the construction of biosafe and biocompatible nanostructures based on nanoparticles of metal oxides and key molecules of the antiviral response in order to increase their activity and improve targeted delivery.
  • Determination of immunomodulatory, antibacterial, anti-inflammatory and hypocholesterolemic properties of probiotic strains of lactobacilli and bifidobacteria, and creation of highly effective new probiotic preparations based on them - immunobiotics. 
  • Development of biotechnological bases for the construction of diagnostic test systems for the identification of pathogens of viral and bacterial infections based on recombinant proteins of human and animal viruses.

The mechanisms of the functioning of interferon and cytokine systems in infectious and non-infectious pathological conditions, the course of which is accompanied by the development of the body's inflammatory reaction, have been determined: infectious diseases of bacterial and viral origin (papillomavirus and herpetic infection of the genitourinary system, vaginitis and dysbiosis of the vagina, herpetic meningoencephalitis), as well as obesity, diabetes Type 2, drug-induced hepatitis, hypertension, etc.

The obtained results made it possible to establish new regularities of the mechanisms of cytokine regulation of the pathogenesis of diseases of various nosological groups, as a result of which the insufficiency of the immune response can be formed at the local and systemic levels, the metabolism of substances (proteins, fats and carbohydrates) can change, the pro- and antioxidant systems and the microbiota of various ecosystems of the body can be disturbed.

Have been determined in preclinical studies Antibacterial, anti-inflammatory, hypocholesterolemic and immunomodulatory properties of probiotic strains of lactobacillus (LAB), bifidobacteria, bacteria species Bacillus subtilis, as well as their separate cellular structures, which can be agonists of receptors involved in providing an immune response to certain pathogen-associated molecular patterns. New probiotic compositions with the maximum level of antibacterial, anti-inflammatory, hypocholesterolemic properties were selected, capable of inducing balancing cytokines (interferons, interleukin-12, -4, -10, etc.), reducing the activity of pro-inflammatory cytokines and balancing the imbalance between Th1/Th2 immune response, as well as normalizing microbiota of different ecosystems of the body in case of one or another level of their violation due to infectious and non-infectious pathological conditions.

On the basis of these studies, scientifically based approaches to the technology of creating the latest bacterial immunotropic drugs are proposed, the main advantages of which are natural origin, targeted immunomodulatory action, complete safety, absence of side effects and complications, the possibility of use for all age groups and use together with chemotherapeutic drugs or other medicines.

A generalized system of knowledge about the role of microbiota in the development of chronic diseases using the example of the metabolic syndrome in the concept of PPDM based on research conducted on own and improved models and in clinical research, new approaches to the translation of the results of biological research are proposed, on the basis of which new methods for treatment and prevention have been created.

The mechanisms of antiviral action of nanobiocomposites based on nanoparticles and cytokines produced by the body in response to the development of infectious and non-infectious processes have been determined. A selection of effective forms and application schemes of the developed nanobiocomposites was carried out, involving both monotherapy and complex use of cytokine cascade inducers and nanobiocomposites, as well as the creation of models for the selection of effective forms and application schemes of the developed nanobiocomposites.

Experimental models have been developed to determine the antibacterial, anti-inflammatory and immunomodulatory properties of probiotic bacterial strains in vivo: models of obesity and diclofenac-induced hepatotoxicity that mimic typical manifestations of the metabolic syndrome. The results of the research became the basis for the further development of recommendations for the prevention of pathological processes of infectious and somatic genesis, the course of which is accompanied by changes in the level of cytokines, carbohydrate and lipid metabolism and microbiota of the gastrointestinal tract and genitourinary system.

The main areas of activity of the Department of Interferon Problems and Immunomodulators have recently included:

  1. creation and research of biosafety, biological activity of nanomaterials based on nanoparticles of metals, oxides of metals and nonmetals;
  2. increasing the effectiveness of vaccination under the influence of probiotic strains of lactobacillus and/or metal nanoparticles;
  3. development of strategic approaches to increase plant virus resistance.
  4. development of the scientific foundations of biotechnology for the production of safe, selenium-containing probiotic preparations and biogenic nanoselenium to increase productivity and preserve poultry stock.

History

The department of problems of interferon and immunomodulators was organized in 1990, with the assistance of Doctor of Science, Professor, Academician of the National Academy of Sciences of Ukraine M. Ya. Spivak. The department employs 11 researchers, including 2 doctors of science and 10 PhDs.

In the department, for the first time, a new scientific direction of interferonology was theoretically substantiated, experimentally proven and clinically confirmed: determination of the antibacterial effectiveness of interferon preparations in bacterial infections (sepsis, purulent-septic processes, salmonellosis, toxic-infectious shock, etc.). The concept of the interferon system was formulated and its direct and reverse connections with the immune and neuroendocrine systems of the body were revealed. A methodology for determining the interferon status of an organism has been developed, which allows establishing the role and place of the interferon system at the population level. By studying the biological features of the cells of the lymphoid organs of the body, the employees of the department developed the biotechnological basis of obtaining interferon preparations from the splenocytes of animals of various species. It has been established that during interferon induction, cells produce other cytokines, primarily tumor necrosis factor, macrophage-activating cytokines, and low-molecular-weight defensin-like peptides with antibacterial activity. This made it possible to develop scientifically based optimal schemes for the use of immunomodulators and to determine their optimal doses proposed for the treatment of patients with various pathological processes (sepsis, radiation injuries, etc.) and to apply new approaches to the prevention and treatment of young farm animals (pigs, cattle, etc.).

The priority results were obtained by elucidating the general problem of induced biosynthesis of α-interferon and designing fundamentally new inducers - molecular complexes of single-chain polyribonucleotides with low molecular weight intercalating ligands. The regularity of the effect of the formation of double-stranded regions in the composition of single-stranded RNA from the structure of a low-molecular-weight ligand was determined. The study of interferon induction under the action of molecular complexes allowed us to propose a new hypothesis regarding the universal mechanism of interferonogenesis, which is reduced to the local deformation of a certain area of ​​the cell membrane with subsequent inclusion of the regulatory system of secondary cellular messengers. Experiments with various inducers of interferon confirmed the theoretical justification.

With the help of modern methods of immunobiotechnology, first- and second-generation interferon preparations, as well as promising interferon inducers, were obtained. Individual indications and contraindications for the clinical use of interferon drugs and its inducers in viral, bacterial diseases and other pathological processes have been determined.

Based on the results of the research, the department added information about the general architecture and morphogenesis of complex animal and plant viruses belonging to one taxon, which is related to the understanding of the fundamental problems of virology in terms of the evolution of phyto- and zoopathogenic rhabdoviruses.

The biosafety and biological activity of nanomaterials based on nanoparticles of metals and nonmetals have been created and investigated. Fundamentally new scientifically based approaches to the construction of biosafe and biocompatible nanostructures based on nanoparticles of metals and drugs have been developed in order to increase their targeted delivery in the case of human pathological conditions. Strategic approaches to increase plant virus resistance have been developed.

The department has a high publishing activity. Employees take an active part in the implementation of international projects, projects, activities of the governing bodies of international scientific organizations. On the basis of the department, a scientific school of Immunobiology and immunomodulators was created, the purpose of which is elucidation of the physiological role of interferon and other cytokines in normal and pathological processes (infectious-inflammatory diseases of bacterial, viral or mixed viral-bacterial origin, as well as in precancerous and tumorous diseases induced by carcinogenic viruses).

The department of problems of interferon and immunomodulators pays considerable attention to establishing contacts with foreign scientific organizations for the purpose of conducting joint research.

Employees of the Department of Interferon and Immunomodulators make great efforts to organize the development of not only fundamental, but also promising applied research on current problems of modern virology, immunology, microbiology and biotechnology, aimed at the implementation of priority areas of development of science and technology, knowledge-intensive technologies, implementation of developments in production. 

Scientific work

The fundamentals of the physiological role of the interferon (IFN) system and other immunoregulatory cytokines in normal and pathological processes: infectious, precancerous, neurodegenerative and other diseases have been determined. It was established that the suppression of IFN-γ production against the background of the tendency to increase the production of Th2-type cytokines - interleukin (IL)-4 and IL-10 and the significant activation of the production of the anti-inflammatory cytokine - transforming growth factor-β1 during the clinically unfavorable course of infectious-inflammatory and preneoplastic cervical diseases (CM) induced by human papillomaviruses (HPV) and herpes simplex viruses (HSV) types 1 and 2, which indicates the activation of inappropriate effector mechanisms of the body's immune defense, can lead to increased susceptibility to viral antigens, and confirms the key role of cytokine profile in the pathogenesis of these diseases. In order to improve the diagnosis, prevention and treatment of patients with PVI, a panel of biomarkers of HPV-induced precancerous and tumorous diseases of the anogenital area has been developed: phenotypic, genotypic, immunological, visual and anamnestic. It has been confirmed that HSV-2 is an important cofactor in the precancerous diseases of CM caused by HPV and can contribute to the growth of HPV-induced tumors. The diagnosis of dysplastic processes of SMH using ultrasonography has been improved.

It has been established that probiotic strains of bacteriaLactobacillus casei IMV B-7280,L. acidophilus IMV B-7279,L. delbrueckii subsp.bulgaricus IMV B-7281,Bifidobscterium animalis VKL and B. animalis VKB in monoculture and in the composition of various probiotic compositions are characterized by a high level of immunomodulatory, as well as antibacterial and anti-inflammatory properties in infectious and inflammatory processes, so they can be used to create highly effective new probiotic preparations - immunobiotics. It has been proven that probiotic strains of lactobacilli can increase the effectiveness of vaccination against hepatitis B, which opens up prospects for improving the clinical effectiveness of vaccines, as well as their biosafety.

The biosafety and biological properties of nanocrystalline cerium dioxide (NCD) have been determined. It has been proven its selective effect on tumor cells, which is manifested in the activation of the production of oxidative stress molecules: reactive oxygen species (ROS) and nitrite ion. A nanobiocomposite based on NDC and IFN was developed, which has a 2-10 times more effective biological effect compared to IFN (separately). It was shown that NDC in the leukocyte-virus system of Newcastle disease exerts a priming effect, ensuring a significant increase in IFN production. Application of IFN-NDC nanobiocomposite in intact animals is accompanied by prolonged activation of the IFN system. Against the background of systemic herpetic infection, the use of NDC for a long time stimulates the IFN system, activates the production of tumor necrosis factor in the early stages of infection, and also modulates the metabolic activity of phagocytic cells. It has been proven that NDC is expedient to use for the modification of the influenza vaccine Vaxigrip, which is confirmed by an increase in its immunogenicity. On the basis of citrate forms of NDC and panthenol, a gel-balm was developed, which was approved for use in the treatment of diabetic patients with diabetic foot syndrome.

Strategic approaches to increase plant virus resistance have been developed. The symptoms of diseases induced by the buckwheat blight virus and their spread have been determined. The harmfulness of this virus has been proven, the sequences of its genomic RNA have been isolated and cloned, the amino acid sequences have been analyzed, and it has also been found reverse correlation between the nuclease activity of buckwheat plants and their sensitivity to this virus. It was shown that transgenic tobacco plants with an expressed human IFN alpha 2b gene showed resistance to infection with buckwheat blight virus. It has been established that buckwheat plants more often have a complex infection induced by viruses, bacteria and microscopic fungi.

During of screening work on bacteria of the genus Lactobacillus based on a detailed study of biological properties two strains were selected Lactobacillus casei IMV B-7280 and L.plantarum IMB B-7679, which are capable of transforming sodium selenite with the formation of biogenic nanoselenium.

The scientific basis for obtaining safe, selenium-containing probiotic preparations capable of transforming sodium selenite into biogenic nanoselenium (Nano-Se) with intracellular and extracellular localization is proposed.

The uniqueness of the department of interferon and immunomodulators problems is related to the determination of the fundamental foundations of the physiological role of the interferon system and other immunoregulatory cytokines under physiological norms and pathological processes, the development of approaches to their correction, the identification of direct and reverse connections of the interferon system with the immune and neuroendocrine systems of the body.

Development of biotechnological aspects of the creation of specific immunobiological preparations of targeted action and diagnostic tools.
Search, study and development of methods of using low molecular weight natural and synthetic substances, nanomaterials for the treatment and prevention of infectious diseases.
For the first time, a fundamentally new direction was proposed and developed - nanotherapy of viral infections, based on the use of cerium dioxide nanoparticles (nanosized substances with a wide range of biological properties) in the formation of antiviral resistance.
In total, he has more than 150 publications, including more than 100 scientific works published in domestic and international peer-reviewed professional publications, including 2 book chapters, 15 patents of Ukraine for utility models and 11 patents for inventions.
